Mycoplasma spp. are common cell culture contaminants that produce and release components into the cell culture environment, including polypeptides, exopolysaccharides and extracellular vesicles, that can alter the cell’s growth properties. This can result in misleading interpretations of the biological processes that are being studied using these cell lines/cultures.
If you suspect mycoplasma contamination or want to be sure your cultures are mycoplasma-free, the Biomedical Research Store is working with the IIHG Genomics Division to provide a mycoplasma detection service. The service uses the LONZA MycoAlert Mycoplasma Detection Assay to screen for the presence of mycoplasma.
Mycoplasma Testing Instructions:
- Grow cells in antibiotic-free media for at least two days.
- Spin down the cells and remove about 1 ml of the supernatant (do not include cells) and put into 1.5 ml microcentrifuge tube.
- Download the Mycoplasma Screen Request Form and complete electronically.
- Bring samples along with the completed form to 116 EMRB (IIHG Genomics Division laboratory). Place samples in refrigerator and the form in the sequence submission box.
- Testing will be completed within 7 days and results emailed. (B/A < 1.0 is interpreted as negative. Borderline B /A ratios (i.e.,1.3) should be retested with a replacement sample).
